Effect of Chlorpromazine and Amytal On the Reduction of Oxidized Glutathione by Rabbit Erythrocytes

Abstract
The reduction of oxidized glutathione by the hemolysate of rabbit erythrocytes was carried out in the presence of glucose, ATP, and NAD or NADP, and the effects of chlorpromazine and Amytal were investigated.
The reduction of oxidized glutathione was much more efficient in the presence of NADP compared with NAD. In the presence of either pyridine coenzyme, chlorpromazine inhibited the reduction of glutathione, while Amytal :showed an enhancing effect. Both compounds showed proportionately greater effectiveness as their concentrations were increased (1-3 mM), such that 3 mM chlorpromazine inhibited the reduction of glutathione by more than 80%, while 3mM Amytal stimulated the reduction more than 2-fold.
These results with rabbit erythrocytes are in a direct contrast to the results obtained by S.S. .Hotta (4) with guinea pig brain mince, and some Horgan specificity in the effect of these drugs on the reduction of glutathione is suggested.
